Analisis Genetik Sifat Ketahanan terhadap Tomato Leaf Curl New Delhi Virus (ToLCNDV) pada Hasil Persilangan Interspesifik Squash dengan Pumpkin

چکیده

Perakitan varietas tahan melalui kegiatan persilangan interspesifik antara Cucurbita maxima (squash) dengan moschata (pumpkin) merupakan upaya untuk mendapatkan squash Tomato Leaf Curl New Delhi Virus (ToLCNDV). Persilangan tersebut menghasilkan individu yang berkarakter dan juga gabungan pumpkin. Pengujian penanda SSR dilakukan guna mengetahui keberhasilan perilangan pada masing-masing tanaman P1 (SQ), P2 (PK), F1 (SQ) (PK). Pool DNA sampel diamplifikasi 10 pasang primer polimorf dapat membedakan pola P1(SQ) (PK) pengujian sebelumnya yaitu CMTm7, CMTm48, CMTm52, CMTm54, CMTm60, CMTm65, CMTm102, CMTp175, CMTp201, CMTp245. Hasil amplifikasi kemudian divisualisasi 2% gel agarosa. menunjukkan bahwa hasil pumpkin (F1) terdapat heterozigot di lokus SSR. Analisis segregasi menggunakan Uji Khi-kuadrat interaksi gen memiliki nisbah 3 Tahan-(AABB, AaBB) : 1 Rentan-(aaBB), mengindikasikan sifat dikendalikan satu dominan penuh. Selain itu gen-gen pengendali ketahanan tetua rentan mengikuti 13 Tahan-(A---, --bb) Rentan-(aaB-), resesif berinteraksi secara epistasis.
